Description(产品描述) 

Soluble native p24 gag protein of Human Immunodeficiency Virus type 1 (IIIB strain). This viral protein was isolated from culture medium of HIV-1 infected MOLT 3 cells, lysed with 0.5% Triton X-100, and purified by immunoaffinity chromatography using mouse monoclonal antibody to HIV-1 p24.

ABI’s Purified Native Viral Proteins are whole viral antigens that have been immunoaffinity purified from clarified infected cell extracts. Recombinant Proteins produced by ABI are specific viral antigens cloned and produced in protein expression systems.

 

ABI offers purified native viral proteins from virus-infected cell culture, isolated from conditioned cell culture medium, inactivated by Triton X-100™, and then purified by immunoaffinity chromatography or reverse phase HPLC to >95% purity. Purified recombinant viral proteins offered by ABI originate from cloned viral genes in Baculovirus, Yeast (Pichia pastoris) or E. coli expression systems and purified by immunoaffinity, ion-exchange, and/or gel exclusion chromatography to >95% purity.

Shipping and Storage: This product is shipped frozen on dry ice. Store at -70°C upon receipt. Avoid multiple freeze-thaw cycles as product degradation may result.

 

Recommendations: Upon thawing, centrifuge the vial for a few seconds to remove residual droplets from the lid.

Safe Handling Recommendation: This biological preparation should be handled in accordance with biosafety guidelines defined in the BMBL, NIH-CDC HHS Publication No. 93-8395.

How are ABI’s Viral Proteins supplied? All ABI proteins are supplied frozen on dry ice.

How are ABI’s Viral Proteins purified? Purification methodology varies according to the individual product requirements, as do the  buffers in which the proteins are frozen.

How pure are ABI’s Viral Proteins?  All of the proteins, whether natural or recombinant, are purified to greater than 95% as  judged by Coomassie Blue stained SDS-PAGE.

How should ABI’s Viral Proteins be stored? We recommend that all proteins be aliquoted into single-use size portions and stored frozen at -70°C or below. Avoid multiple freeze-thaw cycles as this will affect protein activity. Do not dilute  and freeze – make final dilution just prior to use.

How are ABI’s Viral Proteins made? Native proteins are made in infected cells using cell culture. Recombinant proteins are made  in various systems, generally yeast or baculovirus. All are screened for reactivity in ELISA and  Western blot prior to selection.

Will ABI’s Viral Proteins work in ELISA? All ABI proteins work in ELISA.  However, it is critical to use an appropriate primary antibody  and conjugate dilution. Double-block titrations should be performed to determine the optimum  use dilutions in your system.

Will ABI’s Viral Proteins work in Western blot?  All ABI proteins work in Western blot according to the considerations mentioned for ELISA.

I’m getting good Western blot reactivity, but at the wrong molecular weight. What went wrong? It is rare for recombinant proteins to be the same molecular weight as their natural  counterpart. Sequences are either deleted or added as a function of the restriction enzyme sites  in the nucleic acid or the expression vector used to produce the proteins. Additionally, there are  sometimes oligomers and partial cleavage products. Neither affects the function, but sometimes  they will cause confusion. To alleviate doubt, use a monoclonal antibody to demonstrate the  protein in a Western blot or ELISA. Please see the specification sheet for each protein for its  correct molecular weight.

How is the protein concentration of ABI’s Viral Proteins determined? A modified BCA protein assay is used with Bovine Serum Albumin (BSA) as the protein  standard.
